Acute Treatment of Migraine with Oral Rimegepant 75 mg Confers Robust Improvement in Absenteeism, Presenteeism and Productivity: Results from a One Year, Open-Label, Safety Study (BHV3000-201)

To assess the effects of rimegepant on migraine-related absenteeism (ABS), presenteeism (PRE) and lost productivity time (LPT in days).
Recent literature provides compelling data on the severe impact of migraine on daily activity and lost work productivity. Rimegepant is a small molecule CGRP receptor antagonist with demonstrated efficacy and safety in the acute treatment of migraine.
BHV3000-201 was a multicenter, long-term, open-label safety study of rimegepant 75 mg. Eligible subjects included adults with migraine who were instructed to treat migraine attacks of any pain intensity as needed up to once daily for up to 52 weeks. ABS, PRE and LPT were assessed at baseline and weeks 12, 24, 36, and 52 using the validated Migraine Disability Assessment instrument. ABS and PRE were assessed from items 1 and 2 and LPT was derived from the formula, LPT=ABS+PREx0.5. Mean changes from baseline were analyzed post-hoc with paired t-tests, with decreasing scores signifying reduction in lost time (benefit).
Sample sizes were n=1793, 1645, 1217, 1090, and 980 at baseline, weeks 12, 24, 36, 52, respectively. Baseline mean (SE) ABS, PRE, and LPT were 17.7(0.5), 16.2(0.4), and 25.8(0.6) respectively. Mean [95% confidence interval] changes from baseline at weeks 12, 24, 36, 52 were respectively: (1) ABS:−6.7 [−7.5,−5.9]; −7.8 [−8.7,-6.9]; −8.0 [−9.1,-6.9]; −7.9 [−9.0,−6.8]; (2) PRE: −5.9 [−6.6,-5.1]; −6.4 [-7.3.-5.6]; −6.8 [−7.7,-5.9]; −6.9 [−7.8,-5.9] ; (3) LPT: -9.6 [−10.7,−8.5]; −11.0 [−12.2,-9.7]; −11.4 [−12.8,−10.0]; −11.3 [−12.8,−9.9]. Improvements were clinically relevant and statistically significant at all timepoints (p<.0001).
Acute treatment with rimegepant 75 mg confers significant improvements to absenteeism, presenteeism, and improves lost productivity time by 44% (~11 fewer days per month) reflecting improvements in workplace productivity.
